We developed an efficient in vitro propagation system for Haworthia truncata Schönland by testing hormone treatments for primary culture, callus induction, bud differentiation, and rooting. Young, unopened flower buds were the best explants. The optimal initiation medium, consisting of MS with 3.0 mg/L 6-BA and 0.2 mg/L NAA, resulted in callus formation within 7 days. For bud induction, a medium with 1.0 mg/L kinetin achieved a 17-fold callus proliferation rate and 83.7% bud induction after 30 d. Rooting was best on 1/2 MS medium with 1.0 mg/L NAA, with root tips appearing within 10 days, yielding 3.0 taproots per plantlet, averaging 2.0 cm in length after 30 days. These findings show enhanced propagation efficiency of Haworthia truncata tissue culture, addressing issues related to vitrification and leggy growth.
